Recombinant full-length human TPX2 protein was expressed by baculovirus in Sf9 insect cells using an N-terminal His tag. This product can be used for Aurora A activation in vitro.

Scientific Background: TPX2 is a nuclear proliferation-associated protein whose expression is restricted to cell cycle phases S, G2, and M (1). It mainly expressed in the lung carcinoma cell lines and also found in adult placenta, skeletal muscle, thymus, testis, and small intestine and in fetal brain, liver, and kidney.TPX2 is required for targeting STK6 to the spindle apparatus, and that STK6 may regulate the function of TPX2 during spindle assembly. TPX2 is mailnly function in mitotic spindle assembly likely contributes to its role in chromosome stability control and tumor suppression (2).

Uniprot Description

Function: Spindle assembly factor. Required for normal assembly of mitotic spindles. Required for normal assembly of microtubules during apoptosis. Required for chromatin and/or kinetochore dependent microtubule nucleation. Mediates AURKA localization to spindle microtubules. Activates AURKA by promoting its autophosphorylation at 'Thr-288' and protects this residue against dephosphorylation. Ref.13 Ref.17

Subunit structure: Interacts with AURKA. Ref.13 Ref.22 Ref.23

Subcellular location: Nucleus. Cytoplasm › cytoskeleton › spindle. Cytoplasm › cytoskeleton › spindle pole. Note: During mitosis it is strictly associated with the spindle pole and with the mitotic spindle, whereas during S and G2, it is diffusely distributed throughout the nucleus. Is released from the nucleus in apoptotic cells and is detected on apoptotic microtubules. Ref.13 Ref.17

Tissue specificity: Expressed in lung carcinoma cell lines but not in normal lung tissues.

Developmental stage: Exclusively expressed in proliferating cells from the transition G1/S until the end of cytokinesis.

Sequence similarities: Belongs to the TPX2 family.
