Intended Uses: This tPA ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Monkey tPA. This ELISA kit for research use only!

Principle of the Assay: tPA ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-tPA antibody and an tPA-HRP conjugate. The assay sample and buffer are incubated together with tPA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the tPA concentration since tPA from samples and tPA-HRP conjugate compete for the anti-tPA antibody binding site. Since the number of sites is limited, as more sites are occupied by tPA from the sample, fewer sites are left to bind tPA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The tPA concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes tissue-type plasminogen activator, a secreted serine protease which converts the proenzyme plasminogen to plasmin, a fibrinolytic enzyme. Tissue-type plasminogen activator is synthesized as a single chain which is cleaved by plasmin to a two chain disulfide linked protein. This enzyme plays a role in cell migration and tissue remodeling. Increased enzymatic activity causes hyperfibrinolysis, which manifests as excessive bleeding; decreased activity leads to hypofibrinolysis which can result in thrombosis or embolism. Alternative splicing of this gene results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Jul 2008]

PLAT: Converts the abundant, but inactive, zymogen plasminogen to plasmin by hydrolyzing a single Arg-Val bond in plasminogen. By controlling plasmin-mediated proteolysis, it plays an important role in tissue remodeling and degradation, in cell migration and many other physiopathological events. Plays a direct role in facilitating neuronal migration. Increased activity of TPA results in increased fibrinolysis of fibrin blood clots that is associated with excessive bleeding. Defective release of TPA results in hypofibrinolysis that can lead to thrombosis or embolism. Belongs to the peptidase S1 family. 4 isoforms of the human protein are produced by alternative splicing.
