Principle of the Assay: TLR8 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-TLR8 antibody and an TLR8-HRP conjugate. The assay sample and buffer are incubated together with TLR8-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TLR8 concentration since TLR8 from samples and TLR8-HRP conjugate compete for the anti-TLR8 antibody binding site. Since the number of sites is limited, as more sites are occupied by TLR8 from the sample, fewer sites are left to bind TLR8-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TLR8 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the Toll-like receptor (TLR) family which plays a fundamental role in pathogen recognition and activation of innate immunity. TLRs are highly conserved from Drosophila to humans and share structural and functional similarities. They recognize pathogen-associated molecular patterns (PAMPs) that are expressed on infectious agents, and mediate the production of cytokines necessary for the development of effective immunity. The various TLRs exhibit different patterns of expression. This gene is predominantly expressed in lung and peripheral blood leukocytes, and lies in close proximity to another family member, TLR7, on chromosome X. [provided by RefSeq, Jul 2008]
