Porcine Transforming Growth Factor beta ELISA Kit
Principle of the Assay: TGF-beta ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-TGF-beta antibody and an TGF-beta-HRP conjugate. The assay sample and buffer are incubated together with TGF-beta-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TGF-beta concentration since TGF-beta from samples and TGF-beta-HRP conjugate compete for the anti-TGF-beta antibody binding site. Since the number of sites is limited, as more sites are occupied by TGF-beta from the sample, fewer sites are left to bind TGF-beta-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TGF-beta concentration in each sample is interpolated from this standard curve.

This gene encodes an RGD-containing protein that binds to type I, II and IV collagens. The RGD motif is found in many extracellular matrix proteins modulating cell adhesion and serves as a ligand recognition sequence for several integrins. This protein plays a role in cell-collagen interactions and may be involved in endochondrial bone formation in cartilage. The protein is induced by transforming growth factor-beta and acts to inhibit cell adhesion. Mutations in this gene are associated with multiple types of corneal dystrophy.
