Princple of the assay: TF ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-TF antibody and an TF-HRP conjugate. The assay sample and buffer are incubated together with TF-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TF concentration since TF from samples and TF-HRP conjugate compete for the anti-TF antibody binding site. Since the number of sites is limited, as more sites are occupied by TF from the sample, fewer sites are left to bind TF-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TF concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes coagulation factor III which is a cell surface glycoprotein. This factor enables cells to initiate the blood coagulation cascades, and it functions as the high-affinity receptor for the coagulation factor VII. The resulting complex provides a catalytic event that is responsible for initiation of the coagulation protease cascades by specific limited proteolysis. Unlike the other cofactors of these protease cascades, which circulate as nonfunctional precursors, this factor is a potent initiator that is fully functional when expressed on cell surfaces. There are 3 distinct domains of this factor: extracellular, transmembrane, and cytoplasmic. This protein is the only one in the coagulation pathway for which a congenital deficiency has not been described.
