Principle of the assay: TCF20 ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for TCF20. Standards or samples are then added to the microtiter plate wells and TCF20 if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of TCF20 present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for TCF20 are added to each well to “sandwich” the TCF20 immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain TCF20 and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TCF20 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a transcription factor that recognizes the platelet-derived growth factor-responsive element in the matrix metalloproteinase 3 promoter. The encoded protein is thought to be a transcriptional coactivator, enhancing the activity of transcription factors such as JUN and SP1. Alternative splicing results in multiple transcript variants. TCF20: a transcriptional activator. Binds to the regulatory region of MMP3 and thereby controls stromelysin expression. It stimulates the activity of various transcriptional activators such as JUN, SP1, PAX6 and ETS1, suggesting a function as a coactivator. Two alternatively spliced isoforms have been described. Isoform 1 is exclusively expressed in brain, heart and testis, and predominates in liver and kidney. Isoform 2 predominates in lung.
