Specificity
This assay has high sensitivity and excellent specificity for detection of TAS1R3. No significant cross-reactivity or interference between TAS1R3 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between TAS1R3 and all the analogues, therefore, cross reaction may still exist in some cases.

Samples
Serum, plasma, cell culture supernatants, body fluid and tissue homogenate
Assay Type
Quantitative Competitive
Sensitivity
0.1 ng/mL
Preparation and Storage
Store at 2-8 degree C.
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 89% within the expiration date under appropriate storage condition.

Intended Uses: This TAS1R3 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human TAS1R3. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: TAS1R3 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-TAS1R3 antibody and an TAS1R3-HRP conjugate. The assay sample and buffer are incubated together with TAS1R3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TAS1R3 concentration since TAS1R3 from samples and TAS1R3-HRP conjugate compete for the anti-TAS1R3 antibody binding site. Since the number of sites is limited, as more sites are occupied by TAS1R3 from the sample, fewer sites are left to bind TAS1R3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TAS1R3 concentration in each sample is interpolated from this standard curve.

NCBI Description

taste-specific G protein-coupled receptors which recognize natural and synthetic sweeteners [RGD, Feb 2006]

Uniprot Description

Putative taste receptor. TAS1R1/TAS1R3 responds to the umami taste stimulus (the taste of monosodium glutamate) and also to most of the 20 standard L-amino acids, but not to their D-enantiomers or other compounds. TAS1R2/TAS1R3 recognizes diverse natural and synthetic sweeteners. TAS1R3 is essential for the recognition and response to the disaccharide trehalose. Sequence differences within and between species can significantly influence the selectivity and specificity of taste responses ().

Product Notes

The Human TAS1R3 tas1r3 (Catalog #AAA7254769) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7254769 ELISA Kit recognizes Human TAS1R3. It is sometimes possible for the material contained within the vial of "Taste Receptor Type 1 Member 3, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
