RNA Interference (RNAi)
Quality Control
Oligonucleotide synthesis is monitored base by base through trityl analysis to ensure appropriate coupling efficiency. The oligo is subsequently purified by affinity-solid phase extraction. The annealed RNA duplex is further analyzed by mass spectrometry to verify the exact composition of the duplex. Each lot is compared to the previous lot by mass spectrometry to ensure maximum lot-to-lot consistency.
Directions for Use
We recommends transfection with 100 nM siRNA 48 to 72 hours prior to cell lysis. Before resuspending, briefly centrifuge the tube to ensure the lyophilized siRNA is at the bottom of the tube. Resuspend the siRNA oligos to an appropriate concentration with DEPC water. For each vial, suitable for 250 transfections in 24 well plate (20 pmol for each well).
Components
We offer pre-designed sets of 3 different target-specific siRNA oligo duplexes of rat DAB1 gene. Each vial contains 5 nmol of lyophilized siRNA. The duplexes can be transfected individually or pooled together to achieve knockdown of the target gene, which is most commonly assessed by qPCR or western blot. Our siRNA oligos are also chemically modified (2'-OMe) at no extra charge for increased stability and enhanced knockdown in vitro and in vivo.
Preparation and Storage
Shipped at 4 degree C. Store at -20 degree C for one year.

Uniprot Description

DAB1: an adaptor molecule functioning in neural development. The laminar organization of multiple neuronal types in the cerebral cortex is required for normal cognitive function. In mice, the disabled-1 gene plays a central role in brain development, directing the migration of cortical neurons past previously formed neurons to reach their proper layer. Thought to be a signal transducer that interacts with protein kinase pathways to regulate neuronal positioning in the developing brain. Associates with the SH2 domains of Src, Fyn and Abl. Phosphorylated in response to reelin induction in embryonic neurons

Protein type: Adaptor/scaffold

Cellular Component: neuron projection; cell soma; membrane; perinuclear region of cytoplasm; apical part of cell; postsynaptic density; intracellular; cytosol; brush border

Molecular Function: phosphoinositide 3-kinase binding

Biological Process: cell-cell adhesion involved in neuronal-glial interactions involved in cerebral cortex radial glia guided migration; response to drug; cerebellum structural organization; Golgi localization; neuron migration; ventral spinal cord development; negative regulation of cell adhesion; adult walking behavior; cerebral cortex cell migration; positive regulation of protein kinase activity; radial glia guided migration of Purkinje cell; small GTPase mediated signal transduction; negative regulation of astrocyte differentiation; dendrite development; midgut development; negative regulation of axonogenesis; cerebral cortex radially oriented cell migration; brain development; cerebral cortex development; positive regulation of neuron differentiation; negative regulation of JAK-STAT cascade

Product Notes

The DAB1 dab1 (Catalog #AAA8223432) is a siRNA produced from Synthetic and is intended for research purposes only. The product is available for immediate purchase. The DAB1 siRNA (Rat) reacts with Rat and may cross-react with other species as described in the data sheet. AAA Biotech's DAB1 can be used in a range of immunoassay formats including, but not limited to, RNA Interference (RNAi). Researchers should empirically determine the suitability of the DAB1 dab1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "DAB1, siRNA" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
