Quality Control
Oligonucleotide synthesis is monitored base by base through trityl analysis to ensure appropriate coupling efficiency. The oligo is subsequently purified by affinity-solid phase extraction. The annealed RNA duplex is further analyzed by mass spectrometry to verify the exact composition of the duplex. Each lot is compared to the previous lot by mass spectrometry to ensure maximum lot-to-lot consistency.
Directions for Use
We recommends transfection with 100 nM siRNA 48 to 72 hours prior to cell lysis. Before resuspending, briefly centrifuge the tube to ensure the lyophilized siRNA is at the bottom of the tube. Resuspend the siRNA oligos to an appropriate concentration with DEPC water. For each vial, suitable for 250 transfections in 24 well plate (20 pmol for each well).
Components
We offer pre-designed sets of 3 different target-specific siRNA oligo duplexes of mouse GLMN gene. Each vial contains 5 nmol of lyophilized siRNA. The duplexes can be transfected individually or pooled together to achieve knockdown of the target gene, which is most commonly assessed by qPCR or western blot. Our siRNA oligos are also chemically modified (2'-OMe) at no extra charge for increased stability and enhanced knockdown in vitro and in vivo.
Preparation and Storage
Shipped at 4 degree C. Store at -20 degree C for one year.

Uniprot Description

GLMN: Essential for normal development of the vasculature. May represent a naturally occurring ligand of the immunophilins FKBP59 and FKBP12. May function as an membrane anchoring protein. Isoform 1 may stimulate the p70S6K pathway. Isoform 2 may inhibit cell proliferation and increase IL2 production. Defects in GLMN are the cause of glomuvenous malformations (GVMs). GVMs are characterized by the presence of smooth-muscle-like glomus cells in the media surrounding distended vascular lumens. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Cytokine

Cellular Component: Cul2-RING ubiquitin ligase complex; cullin-RING ubiquitin ligase complex

Molecular Function: protein binding; hepatocyte growth factor receptor binding; ubiquitin protein ligase binding; ubiquitin-protein ligase inhibitor activity; receptor binding

Biological Process: positive regulation of interleukin-2 biosynthetic process; regulation of proteasomal ubiquitin-dependent protein catabolic process; regulation of gene expression, epigenetic; negative regulation of cell proliferation; muscle cell differentiation; cell surface receptor linked signal transduction; negative regulation of T cell proliferation; negative regulation of catalytic activity; neural tube closure; negative regulation of protein ubiquitination; vasculogenesis; positive regulation of phosphorylation; positive regulation of cytokine secretion

Product Notes

The GLMN glmn (Catalog #AAA8223731) is a siRNA produced from Synthetic and is intended for research purposes only. The product is available for immediate purchase. The GLMN siRNA (Mouse) reacts with Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's GLMN can be used in a range of immunoassay formats including, but not limited to, RNA Interference (RNAi). Researchers should empirically determine the suitability of the GLMN glmn for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "GLMN, siRNA" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
