RNA Interference (RNAi)
Quality Control
Oligonucleotide synthesis is monitored base by base through trityl analysis to ensure appropriate coupling efficiency. The oligo is subsequently purified by affinity-solid phase extraction. The annealed RNA duplex is further analyzed by mass spectrometry to verify the exact composition of the duplex. Each lot is compared to the previous lot by mass spectrometry to ensure maximum lot-to-lot consistency.
Directions for Use
We recommends transfection with 100 nM siRNA 48 to 72 hours prior to cell lysis. Before resuspending, briefly centrifuge the tube to ensure the lyophilized siRNA is at the bottom of the tube. Resuspend the siRNA oligos to an appropriate concentration with DEPC water. For each vial, suitable for 250 transfections in 24 well plate (20 pmol for each well).
Components
We offer pre-designed sets of 3 different target-specific siRNA oligo duplexes of human NDC80 gene. Each vial contains 5 nmol of lyophilized siRNA. The duplexes can be transfected individually or pooled together to achieve knockdown of the target gene, which is most commonly assessed by qPCR or western blot. Our siRNA oligos are also chemically modified (2'-OMe) at no extra charge for increased stability and enhanced knockdown in vitro and in vivo.
Preparation and Storage
Shipped at 4 degree C. Store at -20 degree C for one year.

NCBI Description

This gene encodes a component of the NDC80 kinetochore complex. The encoded protein consists of an N-terminal microtubule binding domain and a C-terminal coiled-coiled domain that interacts with other components of the complex. This protein functions to organize and stabilize microtubule-kinetochore interactions and is required for proper chromosome segregation. [provided by RefSeq, Oct 2011]

Uniprot Description

HEC1: plays an essential role in chromosome segregation by interacting through its coiled-coil domains with several proteins that modulate the G2/M phase. Phosphorylation by Nek2 is essential for faithful chromosome segregation. Required for the recruitment of Mps1 kinase and Mad1/Mad2 complexes to kinetochores.

Protein type: Cell cycle regulation

Chromosomal Location of Human Ortholog: 18p11.32

Cellular Component: kinetochore; membrane; nucleus; cytosol; chromosome, pericentric region

Molecular Function: protein binding; structural constituent of cytoskeleton

Biological Process: mitosis; mitotic spindle organization and biogenesis; mitotic sister chromatid segregation; attachment of spindle microtubules to kinetochore during mitosis; establishment of mitotic spindle orientation; cell division; mitotic cell cycle; attachment of spindle microtubules to kinetochore; chromosome segregation

Product Notes

The NDC80 ndc80 (Catalog #AAA8223076) is a siRNA produced from Synthetic and is intended for research purposes only. The product is available for immediate purchase. The NDC80 siRNA (Human) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's NDC80 can be used in a range of immunoassay formats including, but not limited to, RNA Interference (RNAi). Researchers should empirically determine the suitability of the NDC80 ndc80 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "NDC80, siRNA" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
