Purity/Purification
Molecular Biology Grade
Endoglycosidase H was purified from the culture fluid of Streptococcus griseus using DEAE-Sephadex and Sephadex G-75.
Form/Format
Supplied as a lyophilized powder from 10mM Tris-HCl, pH7.2 Reconstitute with 0.1ml sterile, distilled, deionized water.
Appearance: Lyophilized powder
Activity
0.1units
Protein: 6ug/vial
Specific Activity:
~30 units/mg protein
Contaminants
No detectable amount of various exoglycosidases or proteases
Quality Control
Endoglycosidase H (0.01 unit) was incubated with appropriate p-nitrophenylglycosides (0.1mg) in 0.2ml of citrate-phosphate buffer (pH 5.0) at 37 degree C for 12 hours. No activites of a- and b-galactosidase or a-L-fucosidase were observed. Absence of contaminating a-mannosidase was also confirmed by using oligosaccharides from ovalbumin and yeast mannan.
Endoglycosidase H (0.01 unit) was incubated with 10mg casein in 1ml of 0.1M Tris-CHl buffer at 37 degree C for 20 hours. 1ml of 10% TCA was added. Release of acid soluble material was below the limit of detection (OD275 less than 0.05).
To inhibit potential protease activity during extended Endo-H digestions, 1mM PMSF and/or 10mM pepstatin A may be added to incubations. PMSF is stable as a 2mM stock solution in absolute methanol.
Unit Definition
One unit is that amount of enzyme required to hydrolyze 1mmole of (Man)5-(GlcNAc)2Asn-[14C]-acetyl, at pH 5.0 and 37 degree C per minute at the saturated substrate concentration. Muramatsu's method is employed).
Preparation and Storage
-20 degree C
Stability: Stable at pH 5.0-9.0 at 37 degree C for 20 hours. Stable for 2 years at -20 degree C. Aliquots of dissolved enzymes may be stored at -20 degree C. Repeated freezing and thawing may decrease the enzyme acitivity.

Product Notes

The Endoglycosidase H (Catalog #AAA653913) is an Enzyme produced from Streptococcus griseus and is intended for research purposes only. The product is available for immediate purchase. It is sometimes possible for the material contained within the vial of "Endoglycosidase H, Enzyme" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
