Principle of the assay: STAT5A ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-STAT5A antibody and an STAT5A-HRP conjugate. The assay sample and buffer are incubated together with STAT5A-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the STAT5A concentration since STAT5A from samples and STAT5A-HRP conjugate compete for the anti-STAT5A antibody binding site. Since the number of sites is limited, as more sites are occupied by STAT5A from the sample, fewer sites are left to bind STAT5A-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The STAT5A concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a member of the STAT family of transcription factors. In response to cytokines and growth factors, STAT family members are phosphorylated by the receptor associated kinases, and then form homo- or heterodimers that translocate to the cell nucleus where they act as transcription activators. This protein is activated by, and mediates the responses of many cell ligands, such as IL2, IL3, IL7 GM-CSF, erythropoietin, thrombopoietin, and different growth hormones. Activation of this protein in myeloma and lymphoma associated with a TEL/JAK2 gene fusion is independent of cell stimulus and has been shown to be essential for tumorigenesis. The mouse counterpart of this gene is found to induce the expression of BCL2L1/BCL-X(L), which suggests the antiapoptotic function of this gene in cells. Alternatively spliced transcript variants have been found for this gene. [provided by RefSeq, Dec 2013]
