Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate SOCS3 in samples. An antibody specific for SOCS3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySOCS3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SOCS3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SOCS3 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a member of the STAT-induced STAT inhibitor (SSI), also known as suppressor of cytokine signaling (SOCS), family. SSI family members are cytokine-inducible negative regulators of cytokine signaling. The expression of this gene is induced by various cytokines, including IL6, IL10, and interferon (IFN)-gamma. The protein encoded by this gene can bind to JAK2 kinase, and inhibit the activity of JAK2 kinase. Studies of the mouse counterpart of this gene suggested the roles of this gene in the negative regulation of fetal liver hematopoiesis, and placental development. [provided by RefSeq, Jul 2008]
