Intended Uses: This SLA ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Canine SLA. This ELISA kit for research use only!

Principle of the Assay: SLA ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-SLA antibody and an SLA-HRP conjugate. The assay sample and buffer are incubated together with SLA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SLA concentration since SLA from samples and SLA-HRP conjugate compete for the anti-SLA antibody binding site. Since the number of sites is limited, as more sites are occupied by SLA from the sample, fewer sites are left to bind SLA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SLA concentration in each sample is interpolated from this standard curve.

NCBI Description

The amino acid selenocysteine is the only amino acid that does not have its own tRNA synthetase. Instead, this amino acid is synthesized on its cognate tRNA in a three step process. The protein encoded by this gene catalyzes the third step in the process, the conversion of O-phosphoseryl-tRNA(Sec) to selenocysteinyl-tRNA(Sec).[provided by RefSeq, Mar 2011]

SLA/LP: Converts O-phosphoseryl-tRNA(Sec) to selenocysteinyl- tRNA(Sec) required for selenoprotein biosynthesis. Defects in SEPSECS are the cause of pontocerebellar hypoplasia type 2D (PCH2D). PCH2D is a disorder characterized by postnatal onset of progressive atrophy of the cerebrum and cerebellum, microcephaly, profound mental retardation, spasticity, and variable seizures. Belongs to the SepSecS family. 3 isoforms of the human protein are produced by alternative splicing.
