Principle of the Assay: HISH3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-HISH3 antibody and an HISH3-HRP conjugate. The assay sample and buffer are incubated together with HISH3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the HISH3 concentration since HISH3 from samples and HISH3-HRP conjugate compete for the anti-HISH3 antibody binding site. Since the number of sites is limited, as more sites are occupied by HISH3 from the sample, fewer sites are left to bind HISH3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The HISH3 concentration in each sample is interpolated from this standard curve.

Variant histone H3 which replaces conventional H3 in a wide range of nucleosomes in active genes. Constitutes the predominant form of histone H3 in non-dividing cells and is incorporated into chromatin independently of DNA synthesis. Deposited at sites of nucleosomal displacement throughout transcribed genes, suggesting that it represents an epigenetic imprint of transcriptionally active chromatin. Nucleosomes wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries which require DNA as a template. Histones thereby play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling.CautionTo ensure consistency between histone entries, we follow the 'Brno' nomenclature for histone modifications, with positions referring to those used in the literature for the 'closest' model organism. Due to slight variations in histone sequences between organisms and to the presence of initiator methionine in UniProtKB/Swiss-Prot sequences, the actual positions of modified amino acids in the sequence generally differ.
