Principle of the assay: SFRP1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-SFRP1 antibody and an SFRP1-HRP conjugate. The assay sample and buffer are incubated together with SFRP1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SFRP1 concentration since SFRP1 from samples and SFRP1-HRP conjugate compete for the anti-SFRP1 antibody binding site. Since the number of sites is limited, as more sites are occupied by SFRP1 from the sample, fewer sites are left to bind SFRP1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SFRP1 concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the SFRP family that contains a cysteine-rich domain homologous to the putative Wnt-binding site of Frizzled proteins. Members of this family act as soluble modulators of Wnt signaling; epigenetic silencing of SFRP genes leads to deregulated activation of the Wnt-pathway which is associated with cancer. They have a role in regulating cell growth and differentiation in specific cell types. SFRP1 decreases intracellular beta-catenin levels. Has antiproliferative effects on vascular cells, in vitro and in vivo, and can induce, in vivo, an angiogenic response. In vascular cell cycle, delays the G1 phase and entry into the S phase.
