Principle of the Assay: SERT ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-SERT antibody and an SERT-HRP conjugate. The assay sample and buffer are incubated together with SERT-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SERT concentration since SERT from samples and SERT-HRP conjugate compete for the anti-SERT antibody binding site. Since the number of sites is limited, as more sites are occupied by SERT from the sample, fewer sites are left to bind SERT-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SERT concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes an integral membrane protein that transports the neurotransmitter serotonin from synaptic spaces into presynaptic neurons. The encoded protein terminates the action of serotonin and recycles it in a sodium-dependent manner. This protein is a target of psychomotor stimulants, such as amphetamines and cocaine, and is a member of the sodium:neurotransmitter symporter family. A repeat length polymorphism in the promoter of this gene has been shown to affect the rate of serotonin uptake and may play a role in sudden infant death syndrome, aggressive behavior in Alzheimer disease patients, and depression-susceptibility in people experiencing emotional trauma.
