Principle of the Assay: The Picokine Human SERPINA6 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Human SERPINA6 with a 96-well strip plate that is pre-coated with antibody specific for SERPINA6. The detection antibody is a biotinylated antibody specific for SERPINA6. The capture antibody is A monoclonal antibody from mouse, the detection antibody is a biotinylated detection polyclonal antibody from goat. The kit contains recombinant Human SERPINA6 with immunogen: Expression system for standard: NSO, Immunogen sequence: M23-V405. The kit is analytically validated with ready to use reagents. To measure Human SERPINA6, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly propotional to Human SERPINA6 in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human SERPINA6 in the sample!!Background/Introduction: Corticosteroid-binding globulin(CBG), also known as Serpin A6 is a protein that in humans is encoded by the SERPINA6 gene. It is mapped to 14q32.13. This gene encodes an alpha-globulin protein with corticosteroid-binding properties. This is the major transport protein for glucorticoids and progestins in the blood of most vertebrates. The gene localizes to a chromosomal region containing several closely related serine protease inhibitors which may have evolved by duplication events.

NCBI Description

This gene belongs to the serpin gene superfamily. Serpins play roles in many processes including inflammation, blood clotting, and cancer metastasis. Members of this family have highly conserved secondary structures with a reactive center loop that interacts with the protease active site to inhibit protease activity. This gene encodes a plasma serine protease that functions as a thrombin and chymotrypsin inhibitor. The protein is activated by heparin, dermatan sulfate, and glycosaminoglycans. Thrombin inhibitor activated by the glycosaminoglycans, heparin or dermatan sulfate. In the presence of the latter, HC-II becomes the predominant thrombin inhibitor in place of antithrombin III (AT-III). Also inhibits chymotrypsin, but in a glycosaminoglycan-independent manner.
