Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-Serpina3n antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-Serpina3n antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Serpina3n amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of Serpina3n can be calculated.

Uniprot Description

SERPINB6: May be involved in the regulation of serine proteinases present in the brain or extravasated from the blood. Inhibitor of cathepsin G, kallikrein-8 and thrombin. May play an important role in the inner ear in the protection against leakage of lysosomal content during stress and loss of this protection results in cell death and sensorineural hearing loss. Defects in SERPINB6 are the cause of deafness autosomal recessive type 91 (DFNB91). It is a form of non- syndromic deafness characterized by progressive and age-dependent sensorineural hearing loss. Vestibular function is normal. Belongs to the serpin family. Ov-serpin subfamily.
