Intended Uses: This immunoassay kit allows for the in vitro quantitative determination of human selenoprotein p,SeP concentrations in serum, Plasma, tissue homogenates and Cell culture supernates and Other biological fluids.

Principle of the Assay||The microtiter plate provided in this kit has been pre-coated with an antibody specific to SeP. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific for SeP and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain SeP, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of SeP in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Description

This gene encodes a selenoprotein containing multiple selenocysteine (Sec) residues, which are encoded by the UGA codon that normally signals translation termination. The 3' UTR of selenoprotein genes have a common stem-loop structure, the sec insertion sequence (SECIS), which is necessary for the recognition of UGA as a Sec codon rather than as a stop signal. This selenoprotein is an extracellular glycoprotein, and is unusual in that it contains 10 Sec residues per polypeptide. It is a heparin-binding protein that appears to be associated with endothelial cells, and has been implicated to function as an antioxidant in the extracellular space. Several transcript variants, encoding either the same or different isoform, have been found for this gene. [provided by RefSeq, Jul 2008]
