Scientific Background: SCYL3 encodes a protein with a kinase domain and four HEAT repeats which interact with the C-terminal domain of ezrin, an ERM protein which play a role in cell adhesion and migration (1). C-terminal domain of SCYL3 interacts with the C-terminal alpha-helical domain of ezrin. Biochemical assays indicate that SCYL3 had associated rather than intrinsic protein kinase activity. SCYL3 is associated with the cytoplasmic face of the Golgi apparatus. This distribution is dependent upon the presence of the SCYL3 N-terminal myristoylation consensus sequence but is not dependent on an association with ezrin. SCYL3 is highly expressed in breast carcinoma cells and adult colon.
NCBI and Uniprot Product Information
NCBI Description
This gene encodes a protein with a kinase domain and four HEAT repeats. The encoded protein interacts with the C-terminal domain of ezrin, an ERM protein, and may play a role in cell adhesion and migration. Alternative splicing results in multiple transcript variants encoding multiple isoforms. [provided by RefSeq, Jun 2012]
Uniprot Description
Function: May play a role in regulating cell adhesion/migration complexes in migrating cells. Ref.1
Subunit structure: Interacts with EZR/VIL2 C-terminal domain. Ref.1
Subcellular location: Cytoplasm. Golgi apparatus. Cell projection › lamellipodium. Note: Colocalized with EZR/VIL2, actin and CD44 in lamellipodia. Ref.1
Tissue specificity: Ubiquitously expressed. Ref.1
Domain: The protein kinase domain is predicted to be catalytically inactive.
Post-translational modification: May be myristoylated; myristoylation may target it to Golgi compartment.Phosphorylated. Ref.1
Sequence similarities: Belongs to the protein kinase superfamily.Contains 4 HEAT repeats.Contains 1 protein kinase domain.

Product Notes
The SCYL3 scyl3 (Catalog #AAA515257) is a Recombinant Protein produced from Sf9 insect cells and is intended for research purposes only. The product is available for immediate purchase. The tag for this protein is GST tag!!Expression System||Sf9 insect cells using baculovirus!!Source Note||Recombinant human SCYL3 (2-end) was expressed by baculovirus in Sf9 insect cells. AAA Biotech's SCYL3 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Researchers should empirically determine the suitability of the SCYL3 scyl3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "SCYL3, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
