Principle of the Assay: The Picokine™ Human RNLS Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Human RNLS with a 96-well strip plate that is pre-coated with antibody specific for RNLS. The detection antibody is a biotinylated antibody specific for RNLS. The capture antibody is a monoclonal antibody from mouse, the detection antibody is a biotinylated polyclonal antibody from goat. The kit contains recombinant Human RNLS with immunogen: Expression system for standard: E.coli, Immunogen sequence: A18-I342. The kit is analytically validated with ready to use reagents. To measure Human RNLS, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly proportional to Human RNLS in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human RNLS in the sample.!!Background/Introduction: Renalase, FAD-dependent amine oxidase is an enzyme that in humans is encoded by the RNLS gene. It is mapped to 10q23.31. Renalase is a flavin adenine dinucleotide-dependent amine oxidase that is secreted into the blood from the kidney. The renalase protein consists of a putative secretory signal peptide (SignalP score of 0.4), a flavin adenine dinucleotide (FAD)-binding region, and an oxidase domain. At least four alternative splicing isoforms have been identified in humans (hRenalase1 to hRenalase4). Only hRenalase1 is detected in human blood samples, which means that hRenalase2 to 4 probably have different functions than hRenalase1.

Renalase is a flavin adenine dinucleotide-dependent amine oxidase that is secreted into the blood from the kidney (Xu et al., 2005 [PubMed 15841207]).[supplied by OMIM, Mar 2008]

RENALASE: Probable FAD-dependent amine oxidase secreted by the kidney, which circulates in blood and modulates cardiac function and systemic blood pressure. Degrades catecholamines such as dopamine, norepinephrine and epinephrine in vitro. Lowers blood pressure in vivo by decreasing cardiac contractility and heart rate and preventing a compensatory increase in peripheral vascular tone, suggesting a causal link to the increased plasma catecholamine and heightened cardiovascular risk. High concentrations of catecholamines activate plasma renalase and promotes its secretion and synthesis. According to PubMed:17385068, is unlikely that renalase has physiologically relevant catecholamine-oxidizing activity. Belongs to the renalase family. 2 isoforms of the human protein are produced by alternative splicing.
