Assay Principle
Typical Laemmli gel systems, which use glycine in the running buffer, are capable of resolving proteins with relative molecular mass no smaller than 10 KD. Very small proteins (<10 KD) are difficult to resolve due to low ability of binding to SDS, which can be solved by gradient gels or using different eletrophoresis conditions, like Tricine-SDS-page. In the tricine system, tricine replaces glycine in the running buffer. This ensures more efficient stacking and destacking of low molecular weight proteins and higher resolution of smaller peptides.
Assay Protocol
1. Mix one volume of Tricine SDS Sample Buffer 2X containing DTT with one volume of protein sample (i. e. add 1mL protein sample into 1mL Tricine SDS Sample Buffer 2X).
2. Heat the sample at 85 degree C for 2 minutes.
3. Load sample into the wells of the SDS-PAGE gel and begin electrophoresis.
4. Run the gel according to the following running conditions.
Voltage: 125V constant
Expected Current: 80mA/gel(start); 40mA/gel(end)
Run time: 90 minutes (dependent on gel percentage)
5. Stop electrophoresis when Coomassie Blue G-250 reaches to the bottom of the gel.

5. Stop electrophoresis when Coomassie Blue G-250 reaches to the bottom of the gel.
General Notes
1. If precipitates or turbidity are observed upon thawing, warm up the solution to 37 degree C for 2-10 min and agitate to dissolve the precipitates.

Upon receipt store at -20 degree C. It is stable for one year. Product is shipped on ice.
