Principle of the Assay: GFI1B ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-GFI1B antibody and an GFI1B-HRP conjugate. The assay sample and buffer are incubated together with GFI1B-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GFI1B concentration since GFI1B from samples and GFI1B-HRP conjugate compete for the anti-GFI1B antibody binding site. Since the number of sites is limited, as more sites are occupied by GFI1B from the sample, fewer sites are left to bind GFI1B-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GFI1B concentration in each sample is interpolated from this standard curve.

This gene encodes a zinc-finger containing transcriptional regulator that is primarily expressed in cells of hematopoietic lineage. The encoded protein complexes with numerous other transcriptional regulatory proteins including GATA-1, runt-related transcription factor 1 and histone deacetylases to control expression of genes involved in the development and maturation of erythrocytes and megakaryocytes. Mutations in this gene are the cause of the autosomal dominant platelet disorder, platelet-type bleeding disorder-17. Alternate splicing results in multiple transcript variants. Alters histone methylation by recruiting histone methyltransferase to target genes promoters. Plays a role in heterochromatin formation. Component of a RCOR-GFI-KDM1A-HDAC complex.
