Principle of the Assay||This kit was based on sandwich enzyme-linked immune-sorbent assay technology. anti-Rat Jak3 antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-Rat Jak3 antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Rat Jak3 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration ofRat Jak3 can be calculated.

NCBI Description

The protein encoded by this gene is a member of the Janus kinase (JAK) family of tyrosine kinases involved in cytokine receptor-mediated intracellular signal transduction. It is predominantly expressed in immune cells and transduces a signal in response to its activation via tyrosine phosphorylation by interleukin receptors. Mutations in this gene are associated with autosomal SCID (severe combined immunodeficiency disease). [provided by RefSeq, Jul 2008]

Mediates signal transduction via the common gamma-chain of cytokine receptors, including IL-2, -4, -7, -9, and -15. Interacts with members of the STAT (signal transduction and activators of transcription) family. Interacts with STAM2 and SHB. Contains two protein kinase domains. Mutations that abrogate JAK3 function cause an autosomal SCID (severe combined immunodeficiency disease). Inhibitor: R017s for organ transplants. Three differentially spliced isoforms have been described.
