Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to Hpt. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific to Hpt. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Hpt, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Hpt in the samples is then determined by comparing the O.D. of the samples to the standard curve.

This gene encodes a histamine H3 receptor that belongs to the superfamily of G-protein coupled receptors. This protein functions as a presynaptic autoreceptor on histamine neurons in the brain, and a presynaptic heteroreceptor in nonhistamine-containing neurons in both the central and peripheral nervous systems. It is deemed a great target for the development of therapeutics for numerous disorders, including obesity, epilepsy, and such cognitive diseases as attention deficit hyperactivity disorder and Alzheimer's disease. Several alternatively spliced transcript variants encoding different isoforms, with different brain expression patterns and signaling properties, have been described for this gene.
