Principle of the Assay: Rat Disrupted In Schizophrenia 1 Protein (DISC1) ELISA Kit employs a two-site sandwich ELISA to quantitate DISC1 in samples. An antibody specific for DISC1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any DISC1 present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Rat DISC1 detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of DISC1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Background/Introduction: Disrupted in schizophrenia 1 is a protein that in humans is encoded by the DISC1 gene. In coordination with a wide array of interacting partners, DISC1 has been shown to participate in the regulation of cell proliferation, differentiation, migration, neuronal axon and dendrite outgrowth, mitochondrial transport, fission and/or fusion, and cell-to-cell adhesion. Several studies have shown that unregulated expression or altered protein structure of DISC1 may predispose individuals to the development of schizophrenia, clinical depression, bipolar disorder, and other psychiatric conditions.
