Principle of the Assay: Rat Cyclooxygenase-1 (COX-1) ELISA Kit employs a two-site sandwich ELISA to quantitate COX-1 in samples. An antibody specific for COX-1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any COX-1 present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Rat COX-1 detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of COX-1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background/Introduction: Cyclooxygenase (COX), first purified in 1976 and cloned in 1988, is the key enzyme in the synthesis of prostaglandins (PGs) from arachidonic acid. The two isoforms of COX are almost identical in structure but have important differences in substrate and inhibitor selectivity and in their intracellular locations. Protective PGs, which preserve the integrity of the stomach lining and maintain normal renal function in a compromised kidney, are synthesized by COX-1. COX-1 is constitutively expressed in most tissues (e.g., platelets, lung, prostate, brain, gastrointestinal tract, kidney, liver and spleen), where the COX-1-derived prostanoids are involved in homeostatic functions.

Uniprot Description

Function: Converts arachidonate to prostaglandin H2 (PGH2), a committed step in prostanoid synthesis. Involved in the constitutive production of prostanoids in particular in the stomach and platelets. In gastric epithelial cells, it is a key step in the generation of prostaglandins, such as prostaglandin E2 (PGE2), which plays an important role in cytoprotection. In platelets, it is involved in the generation of thromboxane A2 (TXA2), which promotes platelet activation and aggregation, vasoconstriction and proliferation of vascular smooth muscle cells

Catalytic activity: Arachidonate + AH2 + 2 O2 = prostaglandin H2 + A + H2O.

Cofactor: Binds 1 heme B (iron-protoporphyrin IX) group per subunit
