Principle of the Assay: CLDN11 ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-CLDN11 antibody and an CLDN11-HRP conjugate. The assay sample and buffer are incubated together with CLDN11-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CLDN11 concentration since CLDN11 from samples and CLDN11-HRP conjugate compete for the anti-CLDN11 antibody binding site. Since the number of sites is limited, as more sites are occupied by CLDN11 from the sample, fewer sites are left to bind CLDN11-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CLDN11 concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the claudin family. Claudins are integral membrane proteins and components of tight junction strands. Tight junction strands serve as a physical barrier to prevent solutes and water from passing freely through the paracellular space between epithelial or endothelial cell sheets, and also play critical roles in maintaining cell polarity and signal transductions. The protein encoded by this gene is a major component of central nervous system (CNS) myelin and plays an important role in regulating proliferation and migration of oligodendrocytes. Mouse studies showed that the gene deficiency results in deafness and loss of the Sertoli cell epithelial phenotype in the testis. This protein is a tight junction protein at the human blood-testis barrier (BTB), and the BTB disruption is related to a dysfunction of this gene.
