Intended Uses: This CALB1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat CALB1. This ELISA kit for research use only!

Principle of the Assay||CALB1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CALB1 antibody and an CALB1-HRP conjugate. The assay sample and buffer are incubated together with CALB1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CALB1 concentration since CALB1 from samples and CALB1-HRP conjugate compete for the anti-CALB1 antibody binding site. Since the number of sites is limited, as more sites are occupied by CALB1 from the sample, fewer sites are left to bind CALB1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CALB1 concentration in each sample is interpolated from this standard curve.

NCBI Description

Calbindin is a calcium-binding protein belonging to the troponin C superfamily. It was originally described as a 27-kD protein induced by vitamin D in the duodenum of the chick. In the brain, its synthesis is independent of vitamin-D-derived hormones. Calbindin contains 4 active calcium-binding domains, and 2 modified domains that presumably have lost their calcium-binding capacity. The neurons in brains of patients with Huntington disease are calbindin-depleted. [provided by RefSeq, Jul 2008]
