Principle of the Assay: SLC25A14 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-SLC25A14 antibody and an SLC25A14-HRP conjugate. The assay sample and buffer are incubated together with SLC25A14-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SLC25A14 concentration since SLC25A14 from samples and SLC25A14-HRP conjugate compete for the anti-SLC25A14 antibody binding site. Since the number of sites is limited, as more sites are occupied by SLC25A14 from the sample, fewer sites are left to bind SLC25A14-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SLC25A14 concentration in each sample is interpolated from this standard curve.

Mitochondrial uncoupling proteins (UCP) are members of the larger family of mitochondrial anion carrier proteins (MACP). Uncoupling proteins separate oxidative phosphorylation from ATP synthesis with energy dissipated as heat, also referred to as the mitochondrial proton leak. Uncoupling proteins facilitate the transfer of anions from the inner to the outer mitochondrial membrane and the return transfer of protons from the outer to the inner mitochondrial membrane. They also reduce the mitochondrial membrane potential in mammalian cells. This gene is widely expressed in many tissues with the greatest abundance in brain and testis. Alternative splicing results in multiple transcript variants. A pseudogene of this gene has been defined on chromosome 4. [provided by RefSeq, Aug 2013]
