Intended Uses: This AchRAb ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse AchRAb. This ELISA kit for research use only!

Principle of the Assay: AchRAb ELISA kit applies the competitive enzyme immunoassay technique utilizing Acetylcholine Receptor antigen and an AchRAb-HRP conjugate. The assay sample and buffer are incubated together with AchRAb-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AchRAb concentration since AchRAb from samples and AchRAb-HRP conjugate compete for the Acetylcholine Receptor antigen binding site. Since the number of sites is limited, as more sites are occupied by AchRAb from the sample, fewer sites are left to bind AchRAb-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AchRAb concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the acyl-CoA thioesterase protein family, and is one of four acyl-CoA hydrolase genes located in a cluster on chromosome 14. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jan 2012]

Uniprot Description

ACOT2: Acyl-CoA thioesterases are a group of enzymes that catalyze the hydrolysis of acyl-CoAs to the free fatty acid and coenzyme A (CoASH), providing the potential to regulate intracellular levels of acyl-CoAs, free fatty acids and CoASH. Displays high levels of activity on medium- and long chain acyl CoAs. Belongs to the C/M/P thioester hydrolase family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Lipid Metabolism - unsaturated fatty acid biosynthesis; Hydrolase; EC 3.1.2.2; Mitochondrial

Chromosomal Location of Human Ortholog: 14q24.3

Cellular Component: mitochondrion

Molecular Function: acyl-CoA hydrolase activity; palmitoyl-CoA hydrolase activity; receptor binding

Biological Process: acyl-CoA metabolic process; very-long-chain fatty acid metabolic process; long-chain fatty acid metabolic process

Product Notes

The Rat ACOT2 acot2 (Catalog #AAA7202446) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7202446 ELISA Kit recognizes Rat ACOT2. It is sometimes possible for the material contained within the vial of "Acyl-coenzyme A thioesterase 2, mitochondrial (ACOT2), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
