This assay has high sensitivity and excellent specificity for detection of ACVR1. No significant cross-reactivity or interference between ACVR1 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between ACVR1 and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This ACVR1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat ACVR1. This ELISA kit for research use only!

Principle of the Assay: ACVR1 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-ACVR1 antibody and an ACVR1-HRP conjugate. The assay sample and buffer are incubated together with ACVR1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ACVR1 concentration since ACVR1 from samples and ACVR1-HRP conjugate compete for the anti-ACVR1 antibody binding site. Since the number of sites is limited, as more sites are occupied by ACVR1 from the sample, fewer sites are left to bind ACVR1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ACVR1 concentration in each sample is interpolated from this standard curve.

NCBI Description

Activins are dimeric growth and differentiation factors which belong to the transforming growth factor-beta (TGF-beta) superfamily of structurally related signaling proteins. Activins signal through a heteromeric complex of receptor serine kinases which include at least two type I ( I and IB) and two type II (II and IIB) receptors. These receptors are all transmembrane proteins, composed of a ligand-binding extracellular domain with cysteine-rich region, a transmembrane domain, and a cytoplasmic domain with predicted serine/threonine specificity. Type I receptors are essential for signaling; and type II receptors are required for binding ligands and for expression of type I receptors. Type I and II receptors form a stable complex after ligand binding, resulting in phosphorylation of type I receptors by type II receptors. This gene encodes activin A type I receptor which signals a particular transcriptional response in concert with activin type II receptors. Mutations in this gene are associated with fibrodysplasia ossificans progressive. [provided by RefSeq, Jul 2008]
