Application Notes
Immunofluroescence: 1:100-1:200 (Fluorescein); 1:1,000-1:2,000 (Streptavidin/HRP)

Tissue Preparation:
10um cryostats
- Fixative: 4% paraformaldehyde in 0.1 M phosphate buffer, pH 7.4; 500 mL over ~20-30 min.
- Post Fixation: 1.5 hr. at 4°C in 4% paraformaldehyde in 0.1 M phosphate buffer, pH 7.4.

Immunofluorescence:
The antibody has significant fluorescein staining at a 1/100-1/200 dilution and significant Biotin-Streptavidin/HRP staining at a 1/2,000-1/4,000 dilution in rat amygdala, and spinal cord. The specificity of the antiserum was evaluated by soluble pre-adsorption with the peptides in question at a final concentration of 10-5 M. CGRP immunolabeling was completely abolished by pre-adsorption with rat α-CGRP and partially eliminated by pre-adsorption with rat β-CGRP. Pre-adsorption with the following peptides resulted in no loss of immunostaining: rat amylin, rat adrenomedulin, calcitonin, neurotensin, somatostatin, substance P, leucine enkephalin, methionine enkephalin, VIP, CCK-8, vasopressin and neuropeptide Y.

Dilutions listed as a recommendation. Optimal dilution should be determined by investigator.

Reconstitution
Do not reconstitute until ready to use since the product is most stable when lyophilized. The product does not need to be cooled during shipping. For long-term storage, store lyophilized antibody until ready to use at -15º C or lower. Reconstitute with 100 uL of distilled or deionized water.
Preparation and Storage
Maintain at 2°C to 8°C for 3 months or at -20°C for longer periods. Stable for up to 1 year. Avoid repeated freeze-thaw cycles.

NCBI Description

This gene encodes the peptide hormones calcitonin, calcitonin gene-related peptide and katacalcin by tissue-specific alternative RNA splicing of the gene transcripts and cleavage of inactive precursor proteins. Calcitonin is involved in calcium regulation and acts to regulate phosphorus metabolism. Calcitonin gene-related peptide functions as a vasodilator and as an antimicrobial peptide while katacalcin is a calcium-lowering peptide. Multiple transcript variants encoding different isoforms have been found for this gene.[provided by RefSeq, Aug 2014]

Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CGRP, Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
