Principle of the Assay: ZPBP2 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-ZPBP2 antibody and an ZPBP2-HRP conjugate. The assay sample and buffer are incubated together with ZPBP2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ZPBP2 concentration since ZPBP2 from samples and ZPBP2-HRP conjugate compete for the anti-ZPBP2 antibody binding site. Since the number of sites is limited, as more sites are occupied by ZPBP2 from the sample, fewer sites are left to bind ZPBP2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ZPBP2 concentration in each sample is interpolated from this standard curve.

Focal adhesions are actin-rich structures that enable cells to adhere to the extracellular matrix and at which protein complexes involved in signal transduction assemble. Zyxin is a zinc-binding phosphoprotein that concentrates at focal adhesions and along the actin cytoskeleton. Zyxin has an N-terminal proline-rich domain and three LIM domains in its C-terminal half. The proline-rich domain may interact with SH3 domains of proteins involved in signal transduction pathways while the LIM domains are likely involved in protein-protein binding. Zyxin may function as a messenger in the signal transduction pathway that mediates adhesion-stimulated changes in gene expression and may modulate the cytoskeletal organization of actin bundles.
