Short Description: Recombinant human FAK protein was expressed in a baculovirus expression system as the full-length protein (accession number NP_722560.1) with an N-terminal DYKDDDDK tag. The molecular weight of the protein is 120.5 kDa. This protein is suitable for use in binding assays, inhibitor screening, and selectivity profiling.

Background: FAK (Focal Adhesion Kinase 1), also known as PTK2 (Protein Tyrosine Kinase 2), is a non-receptor protein-tyrosine kinase that plays an essential role in regulating cell migration, adhesion, spreading, reorganization of the actin cytoskeleton, formation and disassembly of focal adhesions and cell protrusions, cell cycle progression, cell proliferation and apoptosis. It is required for early embryonic development and placenta development, embryonic angiogenesis, normal cardiomyocyte migration and proliferation and normal heart development. FAK regulates axon growth and neuronal cell migration, axon branching and synapse formation, and it is required for normal development of the nervous system. It also plays a role in osteogenesis and differentiation of osteoblasts. FAK can function in integrin signal transduction, but also in signaling downstream of numerous growth factor receptors, G-protein coupled receptors (GPCR), EPHA2, netrin receptors and LDL receptors. It can form multisubunit signaling complexes with SRC and SRC family members upon activation; this leads to the phosphorylation of additional tyrosine residues, creating binding sites for scaffold proteins, effectors and substrates. FAK also can regulate numerous signaling pathways, promote activation of phosphatidylinositol 3-kinase and the AKT1 signaling cascade, MAPK1/ERK2, MAPK3/ERK1 and the MAP kinase signaling cascade. It can recruit the ubiquitin ligase MDM2 to P53/TP53 in the nucleus, and thereby regulates P53/TP53 activity, P53/TP53 ubiquitination and proteasomal degradation.

NCBI Description

This gene encodes a cytoplasmic protein tyrosine kinase which is found concentrated in the focal adhesions that form between cells growing in the presence of extracellular matrix constituents. The encoded protein is a member of the FAK subfamily of protein tyrosine kinases but lacks significant sequence similarity to kinases from other subfamilies. Activation of this gene may be an important early step in cell growth and intracellular signal transduction pathways triggered in response to certain neural peptides or to cell interactions with the extracellular matrix. Several transcript variants encoding different isoforms have been found for this gene, but the full-length natures of only four of them have been determined. [provided by RefSeq, Oct 2015]

Downstream of integrins and Src, upstream of Ras/MAPK. Localizes to focal adhesions that form between cells growing in the presence of extracellular matrix constituents. Interacts with CAS family members and with GIT1, SORBS1 and BCAR3. Interacts with Shb. Required for full Ras transformation of fibroblasts. Increased expression in breast and other cancers, related to chromosome 8q amplification. Overexpression and activation associated with increased migration, invasion and progression of ovarian cancer, and with progression in hepatocellular carcinoma, thyroid cancer, and acute myelogenous leukemia. siRNA increases chemosensitivity of pancreatic adenocarcinoma xenografts. Inhibitor: ISI15421 (antisense). Four splice-variant isoforms have been observed.
