INTENDED USE This PTGIS ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human PTGIS. This ELISA kit for research use only!
PRINCIPLE OF THE ASSAYPTGIS ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PTGIS antibody and an PTGIS-HRP conjugate. The assay sample and buffer are incubated together with PTGIS-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PTGIS concentration since PTGIS from samples and PTGIS-HRP conjugate compete for the anti-PTGIS antibody binding site. Since the number of sites is limited, as more sites are occupied by PTGIS from the sample, fewer sites are left to bind PTGIS-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PTGIS concentration in each sample is interpolated from this standard curve

This gene encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. However, this protein is considered a member of the cytochrome P450 superfamily on the basis of sequence similarity rather than functional similarity. This endoplasmic reticulum membrane protein catalyzes the conversion of prostglandin H2 to prostacyclin (prostaglandin I2), a potent vasodilator and inhibitor of platelet aggregation. An imbalance of prostacyclin and its physiological antagonist thromboxane A2 contribute to the development of myocardial infarction, stroke, and atherosclerosis. [provided by RefSeq, Jul 2008]
