Intended Uses: This PTF1alpha ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Porcine PTF1alpha. This ELISA kit for research use only!

Principle of the Assay: PTF1alpha ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PTF1alpha antibody and an PTF1alpha-HRP conjugate. The assay sample and buffer are incubated together with PTF1alpha-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PTF1alpha concentration since PTF1alpha from samples and PTF1alpha-HRP conjugate compete for the anti-PTF1alpha antibody binding site. Since the number of sites is limited, as more sites are occupied by PTF1alpha from the sample, fewer sites are left to bind PTF1alpha-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PTF1alpha concentration in each sample is interpolated from this standard curve.

This gene encodes a protein that is a component of the pancreas transcription factor 1 complex (PTF1) and is known to have a role in mammalian pancreatic development. The protein plays a role in determining whether cells allocated to the pancreatic buds continue towards pancreatic organogenesis or revert back to duodenal fates. The protein is thought to be involved in the maintenance of exocrine pancreas-specific gene expression including elastase 1 and amylase.
