Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate PSMC5 in samples. An antibody specific for PSMC5 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPSMC5 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PSMC5 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PSMC5 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Uniprot Description

Component of the 26S proteasome, a multiprotein complex involved in the ATP-dependent degradation of ubiquitinated proteins. This complex plays a key role in the maintenance of protein homeostasis by removing misfolded or damaged proteins, which could impair cellular functions, and by removing proteins whose functions are no longer required. Therefore, the proteasome participates in numerous cellular processes, including cell cycle progression, apoptosis, or DNA damage repair. PSMC5 belongs to the heterohexameric ring of AAA (ATPases associated with diverse cellular activities) proteins that unfolds ubiquitinated target proteins that are concurrently translocated into a proteolytic chamber and degraded into peptides.
