Principle of the Assay: PROCR ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PROCR antibody and an PROCR-HRP conjugate. The assay sample and buffer are incubated together with PROCR-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PROCR concentration since PROCR from samples and PROCR-HRP conjugate compete for the anti-PROCR antibody binding site. Since the number of sites is limited, as more sites are occupied by PROCR from the sample, fewer sites are left to bind PROCR-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PROCR concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a receptor for activated protein C, a serine protease activated by and involved in the blood coagulation pathway. The encoded protein is an N-glycosylated type I membrane protein that enhances the activation of protein C. Mutations in this gene have been associated with venous thromboembolism and myocardial infarction, as well as with late fetal loss during pregnancy. Plays a pivotal role in preventing blood coagulation through binding of activated protein C. Expressed strongly in the endothelial cells of arteries and veins in heart and lung, less intensely in capillaries in the lung and skin, and not at all in the endothelium of small vessels of the liver and kidney. The binding of APC to EPCR and PAR-1 increases invasion and chemotaxis.
