Principle of the Assay: PPAR-gamma ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PPAR-gamma antibody and an PPAR-gamma-HRP conjugate. The assay sample and buffer are incubated together with PPAR-gamma-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PPAR-gamma concentration since PPAR-gamma from samples and PPAR-gamma-HRP conjugate compete for the anti-PPAR-gamma antibody binding site. Since the number of sites is limited, as more sites are occupied by PPAR-gamma from the sample, fewer sites are left to bind PPAR-gamma-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PPAR-gamma concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the peroxisome proliferator-activated receptor (PPAR) subfamily of nuclear receptors. PPARs form heterodimers with retinoid X receptors (RXRs) and these heterodimers regulate transcription of various genes. Three subtypes of PPARs are known: PPAR-alpha, PPAR-delta, and PPAR-gamma. The protein encoded by this gene is PPAR-gamma and is a regulator of adipocyte differentiation. Additionally, PPAR-gamma has been implicated in the pathology of numerous diseases including obesity, diabetes, atherosclerosis and cancer. Alternatively spliced transcript variants that encode different isoforms have been described. [provided by RefSeq, Jul 2008]
