Principle of the Assay: PP ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PP antibody and an PP-HRP conjugate. The assay sample and buffer are incubated together with PP-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PP concentration since PP from samples and PP-HRP conjugate compete for the anti-PP antibody binding site. Since the number of sites is limited, as more sites are occupied by PP from the sample, fewer sites are left to bind PP-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PP concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene belongs to the NPY family and it encodes a protein that is synthesized as a 95 aa polypeptide precursor in the pancreatic islets of Langerhans. It is cleaved into two peptide products; the active hormone of 36 aa and an icosapeptide of unknown function. The hormone acts as a regulator of pancreatic and gastrointestinal functions and may be important in the regulation of food intake. Plasma level of this hormone has been shown to be reduced in conditions associated with increased food intake and elevated in anorexia nervosa. In addition, infusion of this hormone in obese rodents has shown to decrease weight gain. [provided by RefSeq, Jul 2008]
