1 unit causes an increase in absorbance at 260 nm of 0.002 per minute per ml at 25ºC when acting upon highly polymerized DNA at pH 4.6.
Note: 0.005 Kunitz units digests 1ug DNA in 10 minutes at 37 degree C, 50mM Tris, 1mM Ca+2, 1mM Mg+2 in a 50ul reaction.
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 6 months after receipt at -20 degree C. For maximum recofery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Related Product Information for DNASE2 enzyme
Deoxyribonuclease II is a glycoprotein endonuclease with a dimeric structure. Deoxyribonuclease II from porcine spleen has a molecular weight of 38kD. Optimum pH range is 4.5-5.0 at ionic strength 0.15M. Deoxyribonuclease II (Acid DNase) hydrolyzes deoxyribonucleotide linkages in native and denatured DNA yielding products with 3'-phosphates. It also acts on p-nitrophenylphosphodiesters at pH 5.6-5.9. Bernardi (BBRC, 17, 573, 1971) describes a three stage degradation of native DNA by DNase II.

Uniprot Description

Function: Hydrolyzes DNA under acidic conditions with a preference for double-stranded DNA. Plays a major role in the degradation of nuclear DNA in cellular apoptosis during development. Necessary for proper fetal development and for definitive erythropoiesis in fetal liver, where it degrades nuclear DNA expelled from erythroid precursor cells

By similarity.

Catalytic activity: Endonucleolytic cleavage to nucleoside 3'-phosphates and 3'-phosphooligonucleotide end-products.

Subcellular location: Lysosome.

Miscellaneous: Not required for the generation of the characteristic DNA fragmentation observed in apoptotic cells, but for the degradation of DNA from dying cells.

Sequence similarities: Belongs to the DNase II family.

Product Notes

The DNASE2 dnase2 (Catalog #AAA653670) is an Enzyme produced from Porcine pancreas and is intended for research purposes only. The product is available for immediate purchase. DNA Sequencing (acidic pH), DNA Digestion (acidic pH). Researchers should empirically determine the suitability of the DNASE2 dnase2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Deoxyribonuclease II, Porcine spleen, Enzyme" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
