Intended Uses: This CTCF ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Porcine CTCF. This ELISA kit for research use only!

Principle of the Assay: CTCF ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CTCF antibody and an CTCF-HRP conjugate. The assay sample and buffer are incubated together with CTCF-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CTCF concentration since CTCF from samples and CTCF-HRP conjugate compete for the anti-CTCF antibody binding site. Since the number of sites is limited, as more sites are occupied by CTCF from the sample, fewer sites are left to bind CTCF-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CTCF concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene is a member of the BORIS + CTCF gene family and encodes a transcriptional regulator protein with 11 highly conserved zinc finger (ZF) domains. This nuclear protein is able to use different combinations of the ZF domains to bind different DNA target sequences and proteins. Depending upon the context of the site, the protein can bind a histone acetyltransferase (HAT)-containing complex and function as a transcriptional activator or bind a histone deacetylase (HDAC)-containing complex and function as a transcriptional repressor. If the protein is bound to a transcriptional insulator element, it can block communication between enhancers and upstream promoters, thereby regulating imprinted expression. Mutations in this gene have been associated with invasive breast cancers, prostate cancers, and Wilms' tumors. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2010]

A critical regulator of cell-cycle arrest and death after B cell receptor signaling in immature B cells. CTCF, together with YY1 and Tsix, form a regulated epigenetic switch for X-inactivation.
