Principle of the Assay: C-MYC ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-C-MYC antibody and an C-MYC-HRP conjugate. The assay sample and buffer are incubated together with C-MYC-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the C-MYC concentration since C-MYC from samples and C-MYC-HRP conjugate compete for the anti-C-MYC antibody binding site. Since the number of sites is limited, as more sites are occupied by C-MYC from the sample, fewer sites are left to bind C-MYC-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The C-MYC concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a multifunctional, nuclear phosphoprotein that plays a role in cell cycle progression, apoptosis and cellular transformation. It functions as a transcription factor that regulates transcription of specific target genes. Mutations, overexpression, rearrangement and translocation of this gene have been associated with a variety of hematopoietic tumors, leukemias and lymphomas, including Burkitt lymphoma. There is evidence to show that alternative translation initiations from an upstream, in-frame non-AUG (CUG) and a downstream AUG start site result in the production of two isoforms with distinct N-termini. The synthesis of non-AUG initiated protein is suppressed in Burkitt's lymphomas, suggesting its importance in the normal function of this gene. [provided by RefSeq, Jul 2008]
