Immunofluorescence (IF)

(Immunofluorescence analysis of ZO-2/TJP2 was done on 90% confluent log phase MCF-7 cells. The cells were fixed with 4% paraformaldehyde for 10 min., permeabilized with 0.1% Triton X-100 for 10 min, and blocked with 1% BSA for 1 hour at RT. The cells were labeled with ZO-2/TJP2 Rabbit Pab at 2ug/mL in 0.1% BSA and incubated for 3 hours at RT and then labeled with Goat anti-Rabbit IgG (H+L) Superclonal Secondary Antibody, Alexa Fluor 488 conjugate at a dilution of 1:2000 for 45 minutes at RT (Panel a: green). Nuclei (Panel b: blue) were stained with SlowFade Gold Antifade Mountant with DAPI. F-actin (Panel c: red) was stained with Alexa Fluor 555 Rhodamine Phalloidin. Panel d is a merged image showing cell junction localization. Panel e is a no primary antibody control. The images were captured at 60X magnification.)

Product Notes

The ZO-2 (Catalog #AAA6506633) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The ZO-2, CT (Tight Junction-associated Polypeptide, Zonula Occludens 2) (Biotin) reacts with Human, Mouse, Rat, Canine and may cross-react with other species as described in the data sheet. AAA Biotech's ZO-2 can be used in a range of immunoassay formats including, but not limited to, Immunofluorescence (IF), Western Blot (WB), Immunocytochemistry (ICC). IF: CaCO2, MDCK II, and NRK52E. Cells need to be fixed with ice-cold ethanol for 30 minutes on ice prior to assays. Applications are based on unconjugated antibody. Researchers should empirically determine the suitability of the ZO-2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "ZO-2, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
