NCBI and Uniprot Product Information
NCBI Description
This gene encodes a member of the Ikaros family of zinc-finger proteins. Three members of this protein family (Ikaros, Aiolos and Helios) are hematopoietic-specific transcription factors involved in the regulation of lymphocyte development. This gene product is a transcription factor that is important in the regulation of B lymphocyte proliferation and differentiation. Both Ikaros and Aiolos can participate in chromatin remodeling. Regulation of gene expression in B lymphocytes by Aiolos is complex as it appears to require the sequential formation of Ikaros homodimers, Ikaros/Aiolos heterodimers, and Aiolos homodimers. Several alternative transcripts encoding different isoforms have been described, as well as some non-protein coding variants. [provided by RefSeq, Apr 2012]
Uniprot Description
Function: Transcription factor that plays an important role in the regulation of lymphocyte differentiation. Plays an essential role in regulation of B-cell differentiation, proliferation and maturation to an effector state. Involved in regulating BCL2 expression and controlling apoptosis in T-cells in an IL2-dependent manner. Ref.6
Subunit structure: Homodimer, and heterodimer with other IKAROS family members. Interacts with IKZF4 AND IKZF5. Interacts with IKZF1. Interacts with HRAS. Interacts with FOXP3; this interaction may be required for silencing target genes and regulating the suppressive activity of FOXP3-positive regulatory T-cells (Treg). Interacts with BCL21L isoform Bcl-X(L); this interaction blocks the anti-apoptotic role of BCL21L. Associates with histone deacetylase complexes containing HDAC1, MTA2 and SIN3A. Ref.3 Ref.6 Ref.7 Ref.8 Ref.9
Subcellular location: Nucleus. Cytoplasm Ref.3 Ref.6.
Tissue specificity: Expressed most strongly in peripheral blood leukocytes, the spleen, and the thymus. Ref.3
Induction: Up-regulated by TGFB1 and 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) in activated AHR T-cells. Ref.9
Post-translational modification: Phosphorylation on tyrosine residues induced by IL2 is required for dissociation from HRAS and nuclear translocation of IKZF3 in T-cells. Phosphorylation on tyrosine residues induced by IL4 is required for dissociation from Bcl-X(L) in T-cells.
Sequence similarities: Belongs to the Ikaros C2H2-type zinc-finger protein family.Contains 6 C2H2-type zinc fingers.

Product Notes
The IKZF3 ikzf3 (Catalog #AAA646760) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The ZNFN1A3 (Zinc Finger Protein Subfamily 1A3, Aiolos, IKAROS Family Zinc Finger 3, IKZF3) reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's ZNFN1A3 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Suitable for use in Western Blot. Dilution: Western Blot: 0.025-1ug/ml. Researchers should empirically determine the suitability of the IKZF3 ikzf3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "ZNFN1A3, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
