Antigen retrieval:
Steam slides in 0.01 M sodium citrate buffer, pH 6.0 at 99-100°C - 20 minutes
Remove from heat and let stand at room temperature in buffer - 20 minutes
Rinse in 1X TBS with Tween (TBST) -1 minute @ RT

Immunostaining:
(Do not allow tissues to dry at any time during the staining procedure)
Apply a universal protein block - 20 minutes @ RT
Drain protein block from slides, apply diluted primary antibody - 45 minutes @ RT

Dehydrate:
(This method should only be used if the chromogen substrate is alcohol insoluble (e.g. Vector Red, DAB)
Wash slides in 2 changes of 80% alcohol - 1 minute each @ RT
Wash slides in 2 changes of 95% alcohol - 1 minute each @ RT
Wash slides in 3 changes of 100% alcohol - 1 minute each @ RT
Wash slides in 3 changes of xylene - 1 minute each @ RT
Apply coverslip
Preparation and Storage
Store at 4°C for up 3 months. Store at -20°C for up to 1 year. Avoid freeze/thaw cycles.

NCBI Description

The protein encoded by this gene is a member of the Zfh1 family of 2-handed zinc finger/homeodomain proteins. It is located in the nucleus and functions as a DNA-binding transcriptional repressor that interacts with activated SMADs. Mutations in this gene are associated with Hirschsprung disease/Mowat-Wilson syndrome. Alternatively spliced transcript variants have been found for this gene.[provided by RefSeq, Jan 2010]

Uniprot Description

ZEB2: Transcriptional inhibitor that binds to DNA sequence 5'- CACCT-3' in different promoters. Represses transcription of E- cadherin. Defects in ZEB2 are the cause of Mowat-Wilson syndrome (MWIS); also known as Hirschsprung disease-mental retardation syndrome. A complex developmental disorder characterized by mental retardation, delayed motor development, epilepsy, microcephaly and a wide spectrum of clinically heterogeneous features suggestive of neurocristopathies at the cephalic, cardiac, and vagal levels. Some patients manifest Hirschsprung disease. Affected patients show an easily recognizable facial appearance with deep set eyes and hypertelorism, medially divergent, broad eyebrows, prominent columella, pointed chin and uplifted, notched ear lobes. Belongs to the delta-EF1/ZFH-1 C2H2-type zinc-finger family.

Protein type: DNA-binding; Motility/polarity/chemotaxis; Transcription, coactivator/corepressor; C2H2-type zinc finger protein

Chromosomal Location of Human Ortholog: 2q22.3

Cellular Component: nucleus

Molecular Function: DNA binding; phosphatase regulator activity; metal ion binding

Biological Process: positive regulation of JNK activity; nervous system development; somitogenesis; transcription, DNA-dependent; cell proliferation in forebrain; hippocampus development; pigmentation during development; positive regulation of melanin biosynthetic process; negative regulation of transcription from RNA polymerase II promoter; positive regulation of melanocyte differentiation; neural tube closure; positive regulation of transcription from RNA polymerase II promoter; neural crest cell migration; positive regulation of Wnt receptor signaling pathway

Disease: Mowat-wilson Syndrome

Product Notes

The ZEB2 zeb2 (Catalog #AAA243094) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Rabbit Polyclonal to Human ZEB2 reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's ZEB2 can be used in a range of immunoassay formats including, but not limited to, Immunohistochemistry (IHC - Paraffin), Immunocytochemistry (ICC), Western Blot (WB), ELISA (EIA). IHC - Paraffin (5ug/ml) Western blot (1-2ug/ml) ZEB2 antibody can be used for detection of ZEB2 by Western blot at 1 - 2 ug/mL. Antibody can also be used for immunocytochemistry starting at 20 ug/mL. For immunofluorescence start at 20 ug/mL. Antibody validated: Western Blot in mouse samples; Immunocytochemistry in human samples and Immunofluorescence in human samples. All other applications and species not yet tested. Western Blot: Predicted: 134 kDa Observed: 135 kDa. Researchers should empirically determine the suitability of the ZEB2 zeb2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "ZEB2, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
