WW domain-containing E3 ubiquitin-protein ligase 1 (WWP1) is a multifunction protein containing an N-terminal C2 domain, four tandem WW domains for substrate binding, and a C-terminal catalytic HECT domain for ubiquitin transferring. WWP1 is a HECT-type ubiquitin E3 ligase that has been implicated in various pathologies. WWP1 is an obesity-inducible E3 ubiquitin ligase that can protect against obesity-associated oxidative stress in WAT. WWP1 which is frequently up-regulated in multiple human malignancies has been demonstrated to play a critical function in cell proliferation, apoptosis, and invasion. Polyubiquitination by the ubiquitin E3 ligase WWP1 suppressed the dimerization, membrane recruitment, and function of PTEN. Either genetic ablation or pharmacological inhibition of WWP1 triggered PTEN reactivation and unleashed tumor-suppressive activity.
