Description: Rabbit IgG polyclonal antibody for WNT1-inducible-signaling pathway protein 1(WISP1) detection. Tested with WB, IHC-P, IHC-F, ICC in Human, Mouse, Rat.
Background: WISP1(WNT1-Inducible Signaling Pathway Protein 1), also known as CCN4, is a matricellular protein that in humans is encoded by the WISP1 gene. WISP1 is induced by WNT1 and belongs to the CCN family, which includes connective tissue growth factor, cysteine-rich-61, and nephroblastoma overexpressed(Tanaka et al., 2001). By use of radiation hybrid mapping panels, Pennica et al.(1998) mapped the WISP1 to chromosome 8q24.1-q24.3, roughly 4 Mb distal to MYC. Pennica et al.(1998) found that 2 distinct systems demonstrated that WISP induction was associated with expression of WNT1. WISP1 genomic DNA was amplified in colon cancer cell lines and in human colon tumors, and its RNA was overexpressed in 84% of tumors examined compared with patient-matched normal mucosa.

NCBI Description

This gene encodes a member of the WNT1 inducible signaling pathway (WISP) protein subfamily, which belongs to the connective tissue growth factor (CTGF) family. WNT1 is a member of a family of cysteine-rich, glycosylated signaling proteins that mediate diverse developmental processes. The CTGF family members are characterized by four conserved cysteine-rich domains: insulin-like growth factor-binding domain, von Willebrand factor type C module, thrombospondin domain and C-terminal cystine knot-like domain. This gene may be downstream in the WNT1 signaling pathway that is relevant to malignant transformation. It is expressed at a high level in fibroblast cells, and overexpressed in colon tumors. The encoded protein binds to decorin and biglycan, two members of a family of small leucine-rich proteoglycans present in the extracellular matrix of connective tissue, and possibly prevents the inhibitory activity of decorin and biglycan in tumor cell proliferation. It also attenuates p53-mediated apoptosis in response to DNA damage through activation of the Akt kinase. It is 83% identical to the mouse protein at the amino acid level. Multiple alternatively spliced transcript variants have been identified. [provided by RefSeq, Mar 2011]

Uniprot Description

WISP1: Downstream regulator in the Wnt/Frizzled-signaling pathway. Associated with cell survival. Attenuates p53-mediated apoptosis in response to DNA damage through activation of AKT kinase. Up-regulates the anti-apoptotic Bcl-X(L) protein. Adheres to skin and melanoma fibroblasts. In vitro binding to skin fibroblasts occurs through the proteoglycans, decorin and biglycan. Belongs to the CCN family. 3 isoforms of the human protein are produced by alternative splicing.

Product Notes

The WISP1 wisp1 (Catalog #AAA176363) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-WISP1 antibody reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's WISP1 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC) Paraffin, Immunohistochemistry (IHC) Formalin, Immunocytochemistry ICC. Researchers should empirically determine the suitability of the WISP1 wisp1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "WISP1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
